Increasing blood glucose and entering glucose into the polyol pathway, is one of the factors involved in the pathogenesis of chronic diabetes complications. Since the key enzyme in this pathway is aldose reductase, the use of plant or chemical compounds that reduce the activity of this enzyme can be used as a therapeutic strategy to prevent the onset of chronic complications of diabetes. In this study, the effect of Gundela tournefortii crude extract and its different fractions in Lorestan province and also several anti-diabetes drugs on inhibition of Aldose reductase enzyme were investigated. For this purpose, firstly, the phenolic and flavonoid content and antioxidant activity of crude extract and various fractions were determined. In the next step, the cow lenses were extracted, the aldose reductase enzyme was isolated by Hayman-Kinoshita method and the inhibitory effect of the compounds on the activity of the enzyme was determined. The results showed that the ethyl acetate fraction has the highest amount of phenol and flavonoid compounds and the highest capacity for the DPPH radical scavenging. Also, this fraction has the highest inhibitory effect on aldose reductase enzyme activity with IC50 = 0.019 mg/ml. This fraction had uncompetitive inhibition with D-glyceraldehyde, but it showed competitive inhibition to NADPH. Anti-diabetic drugs have been shown do not significantly affect the inhibition of the enzyme and are likely to affect other mechanisms of diabetes.}, keywords = {polyol pathway,aldose reductase,chronic diabetes complications,Gundela tournefortii,ethyl acetate fraction}, title_fa = {اثر مهاری فراکسیون موثر گیاه کنگر (Gundelia tournefortii L) و تعدادی از داروهای ضددیابتی بر فعالیت آنزیم آلدوز ردوکتاز لنز چشم: یک مطالعه مقایسه ای}, abstract_fa = {دیابت شیرین در بسیاری از کشورها در سنین 70-20 سالگی علت اصلی کوری، قطع عضو و نارسایی مزمن کلیه محسوب می شود. افزایش سطح گلوکز خون و ورود آن به مسیر پلی‌ال یکی از عوامل دخیل در پاتوژنز عوارض مزمن دیابت است. از آنجایی که آنزیم کلیدی در این مسیر آلدوز ردوکتاز می باشد استفاده از ترکیبات گیاهی یا شیمیایی که فعالیت این آنزیم را کاهش دهند، میتواند به عنوان یک راه درمانی برای جلوگیری از بروز این عوارض مورد استفاده قرار گیرد. در این مطالعه اثر عصاره تام و فراکسیون موثر گیاه کنگر (Gundelai tourneforti) و هم چنین تعدادی از داروهای ضد دیابتی بر روی فعالیت آنزیم آلدوز ردوکتاز مورد بررسی قرار گرفت. برای این منظور، ابتدا محتوای فنول و فلاوونویید و فعالیت آنتی اکسیدانی عصاره تام و فراکسیون های مختلف گیاه تعیین شد. در ادامه لنز چشم گاو استخراج و آنزیم آلدوز ردوکتاز از طریق روش Hayman-Kinoshita جداسازی شد و اثر مهاری ترکیبات مورد نظر روی فعالیت آنزیم مورد بررسی قرار گرفت. دنتایج به دست آمده نشان داد که فراکسیون اتیل‌استاتی دارای بیشترین مقدار ترکیبات فنول و فلاونوئیدی و بالاترین قدرت جمع کنندگی رادیکال های آزاد می باشد. هم چنین، این فراکسیون دارای بالاترین اثرات مهاری روی فعالیت آنزیم آلدوز ردوکتاز با IC50= 0.019 mg/ml می باشد. این فراکسیون با گلیسرآلدهید مهار غیررقابتی و با NADPH مهار رقابتی داشت. مطالعه حاضر روی داروهای ضددیابتی نشان داد که این داروها تاثیر زیادی بر مهار آنزیم ندارند و احتمالا روی مکانیسم‌های دیگر درگیر در بیماری دیابت موثر هستند.}, keywords_fa = {مسیر پلی ال,آلدوز ردوکتاز,عوارض مزمن دیابت,کنگر,فراکسیون اتیل استاتی}, url = {https://animal.ijbio.ir/article_1477.html}, eprint = {https://animal.ijbio.ir/article_1477_11795ed9665cd9740bc963a175240158.pdf} }
