Previous studies have shown that using Zinc Oxide nanoparticles (ZnO-NP) can be a promising agent in cancer treatment, because of their unique properties. The purpose of current study is to investigate the effect of ZnO-NP on oxidative stress in A549 human non-small lung cancer cells. A549 cells with were treated with different concentrations of Zinc Oxide nanoparticles. MTT assay and Trypan blue staining were performed to measure IC50 concentration and cell viability on days 1, 3, 5 and 7. Also, to investigate the apoptotic effects of IC50 concentration of these nanoparticles, Giemsa and AO/EB staining methods and anti-oxidant enzymes superoxide dismutase and catalase activity measurements were used. The results showed that ZnO-NP reduced the A549 cells viabilities in a dose and time dependent manner, and the obtained IC50 concentration was 5mg/mL. The results of stainings indicated an increase in percentage of apoptotic cells in treatment groups. Furthermore, enzyme measurement results showed that the activity of both enzymes in treatment groups was significantly increased in comparison to the control group. The results of this study demonstrated that ZnO-NP have the ability to induce cell death in lung cancer cells and we hope that with organizing this nanoparticle as a new medical compound, it will be used in cancer treatment.}, keywords = {Zinc oxide nanoparticles,Lung cancer,oxidative Stress,Apoptosis}, title_fa = {اثر نانوذرات اکسیدروی بر استرس اکسایشی در سلول های سرطان ریه غیرکوچک انسان A549}, abstract_fa = {سرطان ریه از رایج ترین سرطان ها در مردان و زنان است و در سطح جهانی در حال گسترش می ‌باشد. مطالعات پیشین نشان دادند که استفاده از نانوذرات اکسیدروی به علت خواص منحصر به فردشان، می‌ توانند عامل امیدبخشی در درمان سرطان باشند. هدف از مطالعه حاضر بررسی اثر نانوذرات اکسیدروی بر استرس اکسایشی در سلول های سرطانی غیرکوچک ریه انسان (A549) می‌ باشد. سلول های A549 با غلظت های مختلف از نانوذره اکسیدروی تیمار شد و سنجش MTT و رنگ آمیزی تریپان بلو برای تعیین IC50 و توان زیستی سلول در روزهای 1، 3، 5 و 7 انجام شد. همچنین برای بررسی اثرات آپوپتوزی غلظت IC50 این نانوذرات از روش‌ های رنگ آمیزی گیمسا و اکریدین اورنج/اتیدیوم بروماید و اندازه‌گیری فعالیت آنزیم های آنتی اکسیدانی سوپراکسید دیسموتاز و کاتالاز استفاده‌ شد. نتایج نشان داد که نانوذرات اکسیدروی درصد بقای سلول های A549 را به صورت وابسته به دوز و زمان کاهش داده و غلظت IC50 به دست آمده mg/mL 5 بود. نتایج رنگ ‌آمیزی نشان ‌دهنده افزایش درصد سلول ‌های آپوپتوز یافته در گروه ‌های تیمار سلولی بود. همچنین نتایج آنزیمی ‌نشان داد که فعالیت هر دو آنزیم در گروه ‌های تیمار در مقایسه با گروه کنترل به طور معنی ‌داری افزایش یافت. نتایج این پژوهش نشان داد که نانوذرات اکسیدروی توانایی القای مرگ سلولی را در سلول ‌های سرطانی ریه دارند و امید است که در آینده بتوان با هدفمند کردن این نانوذره به عنوان یک ترکیب دارویی جدید در درمان سرطان مورد استفاده قرار گیرد.}, keywords_fa = {نانوذرات اکسیدروی,سرطان ریه,استرس اکسیداتیو,آپوپتوز}, url = {https://animal.ijbio.ir/article_1728.html}, eprint = {https://animal.ijbio.ir/article_1728_66ef348647aeb30b7cd2610def3bfc7c.pdf} }
