With entering nitrate to the body and converting into other substances it can have detrimental effects on the body and the fetus, and because nitrate passes straight through the barrier placental blood into the liver. The purposes of this study the effects of excess nitrate in drinking water on fetal liver cells of mice were studied. Material and Methods: For this purpose, the effects of the Sodium Nitrate (doses 450, 900 mg ∕ liter/day in drinking water) during pregnancy of female mice were investigated. On 17th day of pregnancy, all groups were killed via chloroform and fetal liver were fixed in 10% formalin for immunohistochemistry and histology analysis. The one–way ANOVA and SPSS software were used to determine the statistical significance of differences between the values for the experimental and control groups and also Image J software were used to cell count. Result: The results obtained showed a significant decrease in weight of pregnant mice, fetal weight, number of fetuses, and also histological changes in liver cells. According to the immunohistochemical study, seems an increase of nitrate in drinking water after entering the liver can be causes BCL2 protein changes in the fetus liver. Conclusion: excess nitrate in drinking water causes disturbances in liver development of the fetus during pregnancy.}, keywords = {Nitrate,Water,Fetal,liver,Immunohistochemistry}, title_fa = {بررسی هیستولوژلوژیکی و ایمونوهیستوشیمیایی اثرات نیترات مازاد در آب آشامیدنی بر روند تکوین کبد جنین موش سوری نژاد NMRI}, abstract_fa = {هدف: امروزه نیترات از طریق کودهای شیمیایی و سوخت های فسیلی وارد آب های زیر زمینی شده و وارد آب آشامیدنی می شود. با توجه به اینکه نیترات از طریق سد خونی جفتی عبور کرده و مستقیما وارد کبد جنین می شود، هدف از این مطالعه اثرات نیترات مازاد در آب آشامیدنی بر سلول های کبدی و اثرات مخرب آن بر روند تکوین بود. مواد و روش ها: در این مطالعه تجربی در روز صفر بارداری موش ها را به سه گروه کنترل، تجربی اول و تجربی دوم (به ترتیب 450 و 900 میلی گرم بر لیتر نیترات در هر روز) تقسیم شدند. در روز 17 بارداری حیوانات بوسیله استنشاق کلروفرم بیهوش کرده و کبد جنین ها جدا و در فرمالین 10% فیکس شد و سپس مورد بررسی ایمونوهیستوشیمی و هیستوپاتولوژی قرار گرفت. در این تحقیق از نرم افزار Image J جهت شمارش سلولی و از نرم افزار SPSS و با آزمون ANOVA یک طرفه جهت آنالیز آماری استفاده شد. نتایج : نتایج حاصل از این مطالعه نشان داد نیترات مازاد در آب تغییرات معناداری در کاهش تعداد و وزن جنین ها، نیز تغییرات هیستوپاتولوژی در سلول های کبدی جنین ایجاد کرد و طبق بررسی ایمونوهیستوشیمی، میزان بیان فاکتور آپوپتوزی BCL2 در بافت کبد در دوره جنینی با افزایش نیترات آب آشامیدنی تغییرات معنی داری نشان داد. نتیجه گیری: نیترات مازاد در آب آشامیدنی سبب اختلالاتی در تکوین کبد جنین در طول دوره بارداری می شود.}, keywords_fa = {نیترات,آب آشامیدنی,جنین,کبد,ایمونوهیستوشیمی}, url = {https://animal.ijbio.ir/article_832.html}, eprint = {https://animal.ijbio.ir/article_832_cb6112d6eefbe667a4f641e53aa0df72.pdf} }
