مطالعه فونستیکی خانواده Cyprinidae از شهرستان پلدختر، ایران
شناسایی ماهیان برای پی بردن به زیست شناسی آن ها و حفاظت از آن ها در برابر تهدید های زیست محیطی یکی از مطالعات مهم می باشد. ماهیان بومی هر منطقه دارای ارزش های متعددی میباشند، این ماهیان جزئی از ذخایر ژنتیکی هر منطقه محسوب میشوند که ما می توانیم از آن ها در تحقیقات ژنتیک و بیوتکنولوژی در آینده استفاده نماییم. در مورد ماهیان مطالعات پراکنده در ایران صورت گرفته است. در پژوژه حاضر، ماهی های آب شیرین شهرستان پلدختر از 8 ایستگاه نمونه برداری در طی سال های1392 تا 1393جمع آوری شدند و پس از نمونه برداری از هر ایستگاه مشخصات جغرافیایی توسط GPS ثبت شد، سپس نمونه ها در فیبرهای مملو از یخ به آزمایشگاه تحقیقاتی دانشگاه اراک انتقال یافتند و مشخصات مورفومتریک هر نمونه اندازه گیری شد و گونه ها توسط کلید های شناسایی معتبر شناسایی شدند. در این مطالعه، 8 گونه متعلق به 6 جنس از خانواده Cyprinidae جمع آوری شدند و گونه های Chondrostoma cyri، Cyprinion tenuiradius، Carassius gibelio، Capoeta damasina، Barbus grypus، Capoeta trutta، Garra rufa، Barbus barbul از شهرستان پلدختر شناسایی شدند.

زیستسنجی جمعیتهای ماهی کولی ارومیه (Alburnus atropatenae)
ماهی کولی ارومیه (Alburnus atropatenae) بومزاد رودخانههای حوضه دریاچه ارومیه است. تعداد 274 قطعه ماهی از هفت رودخانه آقدره، نازلوچای، باراندوز، ساروق، زرینهرود، سنته و تلخهرود مورد بررسی قرار گرفت. تعداد 21 ویژگی اندازشی و 17 ویژگی شمارشی مورد سنجش قرار گرفت. برای تجزیه و تحلیل آماری از آزمونهای پارامتریک و ناپارامتریک و تحلیل متغیرهای کانونی استفاده شد. نتایج تحلیل واریانس یک طرفه ویژگیهای اندازشی نشان داد که 20 ویژگی اندازشی و 14 ویژگی شمارشی به جز تعداد فلسهای پایین خط جانبی، خارهای آبششی و مهرهها، دارای اختلاف معنیدار بودند (05/0p < ) که نشان دهنده وجود تغییرات بالای ریختی در جمعیتهای مورد مطالعه است. تحلیل واریانس یک طرفه، آزمون توکی و دانکن نشان داد که جمعیت تلخهرود در صفات ریختسنجی نسبت به سایر جمعیتها اختلاف معنیداری دارد. در تحلیل مؤلفه اصلی برای صفات ریختسنجی تنها یک مؤلفه اصلی با مقادیر ویژه بزرگتر از 1 شامل 16/84٪ تنوع انتخاب شد. در صفات شمارشی نمودار پراکنش مقادیر اول و دوم نشان داد جمعیت تلخهرود تا حدودی از سایر جمعیتها جدا است. در تجزیه و تحلیل عاملی صفات شمارشی شش مؤلفه اصلی با مقادیر ویژه بزرگتر از یک شامل 496/59٪ تنوع انتخاب شدند. همچنین در صفات شمازشی نمودار پراکنش مقادیر اول و دوم نشان داد جمعیتهای رودخانههای آقدره و نازلوچای همپوشانی داشته ولی از سایر جمعیتها تفکیک شدند. همچنین تلخهرود تا حدودی از جمعیت زرینهرود و به طور کامل از جمعیت ساروق جدا شد. با وجود اختلاف معنیدار بین جمعیتها، تنها ماهیان تلخهرود از سایر جمعیتها تا حدودی جدا شد.

بررسی فعالیت ضد باکتریایی عصاره بدن کرم گلوگاه انار (Zeller) (Lepidoptera: Pyralidae) Ectomyelois ceratoniae بر باکتری Escherichia coli
در سالهای اخیر، بیوتکنولوژیست ها و حشرهشناسان بر مهندسی زیستی پروتئینهای حشرات و کاربرد تولیدات آنها در درمان بیماریها و پزشکی مدرن تأکید فراوانی داشتهاند. مطالعه حاضر با هدف بررسی فعالیت ضد باکتریایی و همولیتیک عصاره بدن لارو Ectomyelois ceratoniae (Zeller) انجام شد. برای تحریک سیستم ایمنی لاروهای کرم گلوگاه انار، سویه های استاندارد قارچ B. bassiana Fashand ، B. thuringiensis var. kurstaki و Escherichia coli به همولنف تزریق شد. فعالیت همولیتیک عصاره القایی و غیرالقایی استخراج شده بعد از 24 ساعت از تزریق بر روی خون تازه انسان، گوسفند و مرغ به دو روش جذب نوری و نشر شعاعی و فعالیت ضد میکروبی به روش نشر شعاعی بر روی باکتری E. coli مورد بررسی قرار گرفت. نتایج نشان داد که عصاره القایی نسبت به عصاره غیر القایی دارای فعالیت ضد میکروبی می باشد. بیشترین فعالیت ضد میکروبی در عصاره القاء شده زیر غشائ فیلتراسیون باکتری E. coli با غلظت 2میلی گرم بر میلیلیتر با میانگین 34/0±7/22 میلیمتر قطر هالههای عدم رشد مشاهده گردید. همچنین بیشترین فعالیت همولیتیک در خون مرغ با میانگین کل 6/39±1/24 درصد در روش جذبی و 25/1±64/5 درصد در روش نشر شعاعی مشاهده گردید. نتایج نشان داد که عصاره بدن لارو کرم گلوگاه انار خاصیت همولیتیکی بسیار کمی بر روی گلبول های قرمز داشت و بسته به نوع عامل پاتوژن القایی فعالیت ضد میکروبی عصاره بر روی باکتری E. coli متفاوت می باشد. با توجه به نتایج این تحقیق، این حشره میتواند کاندیدا خوبی جهت بررسی بیشتری در مطالعات تکمیلی فعالیتهای ضد میکروبی باشد.

بومشناسی لانههای زمستان خوابی خرس قهوهای (Ursus arctos) در ناحیه کوه خم- استان فارس
بخش مهمی از چرخه زندگی خرسهای قهوهای خواب زمستانی است و بنابراین لانه زمستانگذرانی از اهمیت ویژهای برای اینگونه برخوردار است. در این مطالعه ویژگیهای ساختاری لانههای خرسهای قهوهای در منطقه کوهخم در حاشیه شمالی پارک ملی بختگان مورد بررسی قرار گرفت. ویژگیهای لانهها شامل ارتفاع دهانه ورودی، عرض دهانه ورودی، عرض خوابگاه، ارتفاع خوابگاه، طول تونل، عرض تونل، طول لانه و ابعاد بستر خواب در 20 لانه حضور و همچنین 20 غار که آثار حضور خرسها در آنها یافت نشد (نقاط عدم حضور گونه)، ثبت گردید. برای هر لانه متغیرهایی در سطح سیمای سرزمین شامل ارتفاع، شیب، جهت، فاصله تا جاده، فاصله تا سکونتگاههای انسانی و فاصله تا منابع آب نیز اندازهگیری شد. نتایج نشان داد که خرسهای قهوهای در این ناحیه از غارهای طبیعی، شکاف صخرهها و همچنین زیر تختهسنگها برای زمستان خوابی استفاده میکنند. هشت لانه فعال دارای بستر خواب بودند که از مواد گیاهی برای ساخت آنها استفاده شده بود و میانگین طول، عرض و عمق آنها به ترتیب 7 ×70 × 78 سانتیمتر محاسبه شد. همچنین نیمرخ لانههای زمستانگذرانی خرسهای قهوهای این منطقه نیز تعیین و طراحی شد. مقایسه لانههای خرس و مکانهای عدم حضور گونه توسط آزمون t نشان داد که بین متغیرهای ارتفاع و عرض دهانه ورودی در مقیاس خرد و فاصله تا جاده، فاصله تا سکونتگاههای انسانی، فاصله تا منابع آب، شیب و ارتفاع در مقیاس سیمایسرزمین تفاوت معنیداری وجود دارد و احتمالاً این فاکتورها در انتخاب محل خواب زمستانی خرسها مورد توجه قرار میگیرند.

بررسی اثرات سمیتی نانوذرات دی اکسید تیتانیوم بر پارامترهای خونی و سرمی موش سفید آزمایشگاهی
با پیشرفت فناوری نانو و کاربردهای گسترده نانوذرات دی اکسید تیتانیوم در صنایع مختلف، نگرانی در مورد اثرات سمیتی آنها بر محیط زیست و سلامتی انسان وجود دارد. هدف از این مطالعه بررسی اثرات سمیتی نانوذرات دی اکسید تیتانیوم بر پارامترهای خونی و سرمی در موش است. در این مطالعه تجربی، 28 سر موش سوری نر نژاد بالب سی، به چهار گروه شامل یک گروه شاهد و سه گروه آزمایشی تقسیم شدند. نانوذرات دی اکسید تیتانیوم (با غلظتهای 50، 100 و 300 میلی گرم بر کیلوگرم) به مدت 14 روز به صورت دهانی به موشها خورانده شد. در پایان دوره، میزان پارامترهای خونی و سرمی اندازه گیری شدند. نتایج حاصل افزایش قابل توجهی در میزان سلول های سفید خون در گروه دریافت کننده از تمام غلظت های نانوذرات دی اکسید تیتانیوم نسبت به گروه کنترل مشاهده شد ولی در تعداد گلبول های قرمز کاهش مشاهده گردید. میزان هموگلوبین، MCV و نوتروفیلها در غلظت 300 میلی گرم بر کیلوگرم افزایش معنیداری را نشان داد ولی میزان پلاکتها، هماتوکریت، لنفوسیتها، گلوکز و کراتینین به صورت معنی داری کاهش یافتند (P<0.05). یافتهها نشان داد که نانوذرات دی اکسید تیتانیوم سبب اثرات مضری در پارامترهای خونی و سرمی به صورت وابسته به دوز میگردد.

بررسی اکولوژیک شکم پایان منطقه بین جزر و مدی شهرستانهای دیر و کنگان
این مطالعه به منظور شناسایی و تعیین تنوع و تراکم شکم پایان منطقه بین جزرومدی سواحل شهرستان های دیر و کنگان درسال 1393 از 8 ایستگاه در دو فصل بهار وپاییز نمونه برداری شد .نمونه برداری از مناطق بین جزرومدی با استفاده از کوادرات cm50×50 با سه تکرار در هر ایستگاه صورت گرفت. نمونه ها پس از جمع آوری در فرمالین % 4 فیکس و به آزمایشگاه منتقل و مورد شمارش و شناسایی قرار گرفت. خصوصیات فیزیکو شیمیایی آب از قبیل دما، pH ، اکسیژن محلول و شوری نیز در هر ایستگاه اندازه گیری شد. در این بررسی مجموعاً 19 گونه متعلق به 10 خانواده از شکم پایان مورد شناسایی قرار گرفتند. بیشترین گونه های شناسایی شده در ایستگاههای مورد مطالعه از خانواده هایNeritidae ،Trochidae، Muricidae و Acmaeidae بودند. شاخص شانون(H´) و شاخص سیمسون (λ)در بین فصول نمونه برداری و ایستگاه های نمونه برداری دارای اختلاف معنی داری می باشند اما شاخص مارگالف(R) تنها بین ایستگاههای نمونه برداری دارای اختلاف معنی دار می باشد(05/0 P<)و در بین فصول نمونه برداری دارای اختلاف معنی دار نمی باشد)05/0(P>. با توجه به گونه های شناسایی شده و همچنین شاخص کیفیت آب )ولچ) می توان بیان نمود که سواحل مورد مطالعه دارای آب با کیفیت متوسط و تقریباً ضعیف میباشند.

تولید ژله رویال، مقدار تیامین ژله رویال و توسعه غدد هیپوفارنژیال در کلنیهای زنبور عسل ایرانی (Apis mellifera Meda) تغذیه شده با سطوح مختلف تیامین
برای بررسی اثر تغذیهای تیامین (ویتامین1B) بر توسعه غدد هیپوفارنژیال، تولید و میزان تیامین ژله رویال کلنی زنبورعسل ایرانی (Apis mellifera M.) آزمایشی در شرایط طبیعی در مؤسسه تحقیقات علوم دامی کشور انجام شد. در این آزمایش 20 کلنی زنبور عسل ایرانی در پنج گروه تغذیهای با چهار تکرار مورد استفاده قرار گرفت. کلنیهای گروه کنترل با شربت شکر 1:1 و کلنیهای آزمایشی به روش مشابه ولی با این تفاوت تغذیه شدند که شربت شکر با سطوح مختلف تیامین (100، 200، 300 و 400 پیپیام) مکملسازی شده بود. عملیات صحرایی به مدت60 روز (30 روز تغذیه با شربت و 30 روز بعد از تغذیه) انجام گرفت. برای بررسی رشد و نمو غدد هیپوفارنژیال، طول و عرض ده آسینی برای هر زنبور کارگر در 3، 6، 9، 12 و 15 روزگی با استفاده از استریومیکروسکوپ اندازهگیری شد. همچنین برای تعیین تیامین ژله رویال یک فرایند استخراج بهوسیله دستگاه کروماتوگرافی (HPLC) انجام شد. نتایج آزمایش نشان داد که سطح آسینی غدد هیپوفارنژیال زنبورهای کارگر در کلنیهای تغذیه شده با سطوح مختلف تیامین دارای اختلاف معنیداری میباشد. زنبورهای تغذیه شده با سطوح بالای تیامین سطح آسینی بیشتری داشتند (05/0P<). تجزیه محتوای تیامین نمونههای ژله رویال با استفاده از دستگاه HPLC نشان داد که مقدار تیامین در ژله رویال کلنیهای تغذیه شده با 400 پیپیام تیامین بیشتر از (7/2 میلیگرم در صد گرم) کلنیهای دیگر بود (05/0P<). به طور کلی میتوان گفت که استفاده از سطوح مختلف تیامین باعث بهبود عملکرد کلنیهای زنبورعسل میشود.

اثر استرس شوری در بیان ژن Na/KATPase در دو جمعیت مختلف آرتمیا Artemia franciscana و Parthenogenetic Artemia با تکنیک RT-PCR-DGGE
آرتمیا یکی از انواع سخت پوستان است که شوریهای بسیار بالا را تحمل می کند. به واسطهی تغییرات اقلیمی و بالا رفتن شوری منابع آبهای داخلی و تالاب ها و همچنین اهمیت آرتمیا به عنوان یکی از فاکتورهای مهم در امر پرورش و تغذیه لارو ماهی و میگو، بررسی ویژگیهای مولکولی جمعیتهای مختلف آرتمیا ضروری به نظر میرسد. مطالعه حاضر با هدف تاثیر استرس شوری بر روی بیان ژن Na+/K+ATPase در اگزون شماره 7 دو جمعیت مختلف آرتمیا با استفاده از توسط تکنیک RT-PCR-DGGE انجام شد. در این تحقیق ابتدا 2 جمعیت از آرتمیاهای گونه Artemia franciscana و Parthenogenetic Artemia در شوریهای 60، 120 و 180 گرم در لیتر پرورش داده شد. در انتهای دوره mRNA ژن Na+/K ATPase تهیه و پس از تولید cDNA، تصویر آن با استفاده از تکنیک DGGE بررسی و همچنین باندهای ایجاد شده توالییابی شدند. نتایج این بررسی نشان داد محصول تولید شده از این ژن در A. franciscana هموزیگوت، در حالیکه در Parthenogenetic Artemia هتروزیگوت که حاصل دو الل است. همچنین در Parthenogenetic Artemia حاصل ترجمه پروتئینی که از دو آلل مختلف حاصل شده است. جالب اینکه بر اساس نتایج این مطالعه، هیچ تفاوتی در نمونههای مورد بررسی در شوریهای مختلف مشاهده نشد و ظاهراً تنها اختلاف موجود در ژنوم این جمعیتها میباشد.

مطالعه میگوی سفید سرتیز (Metapenaeus affinis ) سواحل بوشهر از نظر آلودگی به فلزات سنگین آهن، روی، مس ، منگنز و نیکل و تخمین ریسک برای مصرف کننده
اکوسیستمهای دریایی از طریق تخلیه صنعتی، رواناب کشاورزی و شهری و زباله های شهری به فلزات سنگین آلوده می-شوند. بنابراین موجودات آبزی این آبها، احتمالا آلوده به فلزات سنگین بوده و برای مصرف ایمن نیستند. این تحقیق در سال 1395 با هدف تعیین غلظت فلزات سنگین) آهن، روی، مس، منگنز و نیکل (در بافت خوراکی میگو سفید سرتیز سواحل بوشهر و ارزیابی خطر غذایی ناشی از مصرف آنها انجام شد. نمونههای میگو از سواحل بوشهر به روش تصادفی و با استفاده از قایق و تور صیادی جمع آوری شد. از روش هضم اسیدی جهت آماده سازی نمونهها استفاده شد و غلظت فلزات سنگین توسط دستگاه اسپکتروسکوپی جذب اتمی شعلهای (FAAS)تعیین شد. برای تجزیه و تحلیل دادهها از نرم افزار آماری 19SPSS استفاده گردید. برآورد ﺟﺬب روزاﻧﻪ و ﭘﺘﺎﻧﺴﻴﻞ ﺧﻄﺮ فلزات سنگین ﻧﺎﺷﻲ از ﻣﺼﺮف اﻳﻦ گونه میگو بر اساس دستور کار EPA انجام ﺷﺪ. در این تحقیق میانگین غلظت فلزات آهن، روی، مس، منگنز و نیکل در بافت عضله میگو به ترتیب 05/85±92/130، 00/11±61/65، 29/34±81/35، 22/4±20/10، 49/7±66/10میلیگرم بر کیلوگرم وزن خشک به دست آمد. بر اساس یافتهها، غلظت فلزات آهن، روی و مس در مقایسه با استانداردهای جهانی پایین تر بود. میزان فلز نیکل به طور معنیداری در مقایسه با استانداردFDA بالاتر اما در مقایسه با سایر استانداردها پایینتر بود. میزان جذب روزانه و هفتگی پایین تر از دوز مرجع EPA بود. در شرایط حاضر، گونه میگوی مورد مطالعه برای مصرف ایمن است و مصرف مداوم آن (70 سال) سبب ایجاد اثرات غیر سرطان زا نمی شود.

تاثیرات حمایتی عصاره هیدرواتانولی میوهی گیاه نسترن کوهی در مسمومیت کبدی و کلیوی القا شده با اتانول
مقدمه: گیاه نسترن کوهی به شکل گسترده ایی در طب سنتی ایرانی مورد استفاده قرار میگیرد. این مطالعه جهت بررسی فعالیت حمایتی نسترن کوهی در مسمومیت کبدی القا شده با اتانول در رَتها، طراحی شد. مواد و روشها: در مطالعهی حاضر، 30 سر رَت ویستار به 5 گروه تقسیم شدند شامل: کنترل، اتانول (5 g. kg−1)، اتانول+عصارهی 1(200 mg. kg−1) و اتانول+عصارهی 2(400 mg. kg−1) و عصاره تنها (400 mg. kg−1). بررسی هیستوپاتولوژیکی، اندازهگیری آسپارتات آمینوترانسفراز (AST)، آلانین آمینوترانسفراز (ALT)، آلکالین فسفاتاز (ALP) و سطح اوره (BUN) و کراتینین (Creatinine) سرم خون صورت گرفت. نتایج: مصرف اتانول منجر به افزایش معنیداری در سطح سرمی AST، ALT، ALP و اوره و کراتینین میشود. تیمار همزمان عصارههای دوزهای مختلف نسترن کوهی به همراه اتانول، به شکل معنی داری سطوح AST، ALT، ALP، اوره و کراتینین را کاهش میدهد. همچنین عصارهی با دوزهای مختلف منجر به کاهش آسیبهای بافتی می-گردد. نتیجهگیری: یافتههای ما پیشنهاد میکند که گیاه نسترن کوهی توانایی حمایت از کبد و کلیه در برابر تغییرات بیوشیمیایی و هیستوپاتولوژیکی القا شده با اتانول را دارا میباشد.
