@article { author = {}, title = {A laboratory investigation on the effect of environmental temperature on virulence of Helicoverpa armigera Multiple Nucleopolyhedrovirus}, journal = {Journal of Animal Research (Iranian Journal of Biology)}, volume = {26}, number = {4}, pages = {372-383}, year = {2014}, publisher = {Iranian Biology Society}, issn = {2383-2614}, eissn = {2383-2622}, doi = {2643}, abstract = {Not only a low speed of the effect of virus is one of factors to limit the use of baculoviruses in the field but also microclimate thermal condition is a lateral factor besides other effective factors that can affect on virulence and speed of the effect of virus.  In this evaluation the effect of different temperatures 20-30 °C were investigated in biological activity (LT50, LD50) of a Helicoverpa armigera Multiple Nucleopolyhedrovirus isolate in early 3 rd instar larva of cotton budworm ( Helicoverpa armigera Hub.) in laboratory condition (50-60% relative humidity and 16L:8D photoperiod) in a factorial split plot. The viral dosage and the required time for mortality were similarly important in the temperature factor.  LD50 values in temperatures 20, 26 and 30 °C were estimated 3580.83, 2614.82 and 1751.62 PIB/ Larvae respectively.  The comparison of the mean LD50 values with Duncan,s multiple range test in three temperatures showed that viral range of mortality increases  significantly with increasing temperature.  The effect of temperature in mortality is especially significant in low dosages.  LT50 values in viral dosage of 5 ×10 5 PIB/ Larvae in temperatures 20, 26 and 30 °C were estimated 7.15, 6.73 and 6.58 day respectively.  The comparison of the mean LT50values with Duncan,s multiple range test show that viral dosage in 30 °C is more active than 20 °C and its activity in 26 °C isn’t significantly different from two other temperatures.  Evaluation of LD50 and LT50 values of viral dosage and its comparison to incubation period show that the effect of temperature in the mortality rate and mortality speed is after the incubation period.  Therefore temperatures in range of 26-30 °C could be recommended in reaching the high mortality percent and increasing the viral speed mortality.}, keywords = {Helicoverpa armigera Multiple Nucleopolyhedrovirus,environmental temperature,incubation period,Helicoverpa armigera,LD50 and LT50}, title_fa = {بررسی تاثیر دمای محیط در قدرت بیماریزایی ویروس چند وجهی هسته‌ای کرم غوزه پنبه Helicoverpa armigera multiple nucleopolyhedrovirus در آزمایشگاه}, abstract_fa = {پایین بودن سرعت تاثیر ویروس در مزرعه یکی از عوامل محدودکننده کاربرد باکولوویروس‌ها می‌باشد. دمای حاکم بر شرایط میکروکلیمایی موجود در منطقه از جمله عواملی است که به عنوان یک عامل جانبی در کنار سایر عوامل موثر در کارایی و سرعت تاثیر ویروس اثرگذار می‌باشد. در این بررسی تاثیر دمای 30- 20 درجه سانتی‌گراد بر فعالیت بیولوژیکی(LD50 و LT50) ویروس چند وجهی هسته‌ای کرم غوزه پنبه (Helicoverpa armigera multiple nucleopolyhedrovirus) در لاروهای اوایل سن 3 کرم غوزه پنبه  Helicoverpa armigera Hub.در شرایط نوری D8 : L16 و رطوبت نسبی 60- 50% در قالب طرح اسپلیت پلات فاکتوریل به صورت دز و زمان لازم برای مرگ و میر با اهمیت برابر در داخل عامل دما مورد ارزیابی قرار گرفت.  مقادیر LD50 مربوط به ویروس در دماهای 20، 26 و 30 درجه سانتی‌گراد به ترتیب 83/3580، 82/2614 و 62/1751 پلی‌هدر در هر لارو برآورد شد.  مقایسه میانگین مقادیر LD50 با آزمون دانکن در هر سه دما نشان می‌دهد که با افزایش دما میزان مرگ و میر حاصل از ویروس به طور معنی‌دار افزایش می‌یابد.  تاثیر دما در میزان مرگ ومیر به ویژه در دزهای پایین معنی‌دار می‌باشد.  مقادیر LT50 مربوط به دز 105 × 5 پلی‌هدر در هر لارو در دماهای 20، 26 و 30 درجه سانتی‌گراد به ترتیب 15/7، 73/6 و 58/6 روز برآورد شد.  مقایسه میانگین مقادیر LT50 با آزمون دانکن نشان می‌دهد که ویروس در دمای 30 درجه سانتی‌گراد سریع‌تر از 20 درجه سانتی‌گراد عمل کرده و در دمای 26 درجه سانتی‌گراد اختلاف معنی‌داری با دو دمای دیگر ندارد.  بررسی مقادیر LD50 و LT50 مربوط به ویروس و مقایسه آن با دوره نهفته آلودگی نشان می‌دهد که تاثیر دما در سرعت و میزان مرگ ومیر بعد از دوره نهفته آلودگی می‌باشد. لذا دماهای بالای 30- 26 درجه سانتی‌گراد جهت حصول درصد مرگ ومیر بیشتر و افزایش سرعت مرگ ومیر ناشی از ویروس توصیه می‌شود.}, keywords_fa = {ویروس چند وجهی هسته‌ای کرم غوزه پنبه,دمای محیط,دوره نهفته آلودگی,کرم غوزه پنبه,Helicoverpa armigera,LD50 و LT50}, url = {https://animal.ijbio.ir/article_258.html}, eprint = {https://animal.ijbio.ir/article_258_dff9ba9b9e942ff0282b88777fca40f4.pdf} }
