چکیده
زمینه و هدف: تنش حرارتی میتواند منجر به تنش اکسیداتیو شود، که در نتیجه وضعیت ایمنی، سلامتی و عملکرد سلولهای موجود زنده را تحت تاثیر قرار میدهد. از طرفی خواص آنتیاکسیدانی اسید آلفا لیپوئیک و نقش کروم در متابولیسم گلوکز با کاهش اثرات ناشی از تنش اکسیداتیو همراه است.
مواد و روشها: در مطالعه حاضر BMSCs از جوجه یکروزه نژاد راس 308 جدا شدند. برای تمایز BMSCs به سلولهای چربی و استخوانی به ترتیب با محیط آدیپوژنیک و استئوژنیک تیمار شدند. گروههای آزمایشی شامل: سلولهای بدون هیچ گونه تیماری، سلولهایی که دچار شوک حرارتی شدند، سلولهای تحت شوک حرارتی تیمارشده با غلطت 100 میکرومولار پروپیونات کروم و 100 میکرومولار اسید آلفا لیپوئیک به صورت جداگانه و همزمان. سـپس، BMSCs از لحاظ مورفولوژی، بقا، سنجش فاکتورهای آنتیاکسیدان و میزان بیان ژنهای مرتبط با ایمنی مقایـسه شـدند. تجزیه وتحلیل دادهها نیز با استفاده از نرم افزار SAS انجام گرفت.
یافتهها: سلولهای جدا شده از نظر ریختشناسی، فیبروبلاستی بودند و با استفاده از فلوسایتومتری و پتانسیل تمایزی تعیین هویت شدند. بیشترین میزان فعالیت آنزیمهای SOD، CAT، GPX و TAC در گروه اسید آلفا لیپوئیک تحت شوک حرارتی مشاهده شد. کمترین میزان بیان ژن IL-2، IL-6 و INF-γ به ترتیب در گروه سلولهای تیمار شده همزمان با پروپیونات کروم و اسید آلفا لیپوئیک و گروه سلولهای تیمار شده اسید آلفا لیپوئیک مشاهده شد.
نتیجهگیری: نتایج نشان داد که استفاده از مکملهای آنتیاکسیدان در شرایط شوک حرارتی سبب بهبود پاسخهای آنتیاکسیدانی و ایمنی میشود.

Background and Objective: Heat stress can lead to oxidative stress, as a result, it affects the immune status, health, and function of living cells. On the other hand, the antioxidant properties of alpha-lipoic acid and the role of chromium in glucose metabolism are associated with reducing the effects of oxidative stress. Tantioxidant status and the immune response of cultured cells under heat stress.
Materials and Methods: In the present study, BMSCs were isolated from day-old chicks of Ras 308 breed. To differentiate BMSCs into fat and bone cells, they were treated with adipogenic and osteogenic medium, respectively. Experimental groups include cells without any treatment, cells that suffered heat shock, and heat-shocked cells treated with 100 μM chromium propionate and 100 μM alpha-lipoic acid separately and simultaneously. Then, BMSCs were compared in terms of morphology, survival, measurement of antioxidant factors, and the expression level of genes related to immunity. Data were analyzed through SAS software.
Results: The isolated cells were morphologically fibroblastic and identified using flow cytometry and differentiation potential. The highest level of activity of SOD, CAT, GPX, and TAC enzymes was observed in the alpha-lipoic acid group under heat shock. The lowest levels of IL-2, IL-6, and INF-γ gene expression were observed in the group of cells treated simultaneously with chromium propionate and alpha-lipoic acid and in the group of cells treated with alpha-lipoic acid, respectively.
Conclusion: The results showed that the use of antioxidant supplements in heat shock conditions improves antioxidant and immune responses.
