چکیده

ساختمان آناتومیکی چشم، نوع غدد اشکی و ترشحات آن در عدم بروز بعضی از آسیب های وارده به چشم می تواند مهم باشد. در این تحقیق از پنج عدد عروس هلندی نر سالم استفاده شد. پس از تشریح نمونه ها و تعیین موقعیت قرار گرفتن غده، با استفاده از روش های معمول آماده سازی بافت مقاطع بافتی تهیه شد وبا هماتوکسیلین - ائوزین، پاس، آلسین بلو و تولوئیدین بلو رنگ آمیزی گردیدند.نتایج نشان داد که غدۀ هاردرین در عروس هلندی مثلثی شکل بوده و در سطح شکمی داخلی کرۀ چشم قرار دارد. میانگین طول آن5/4 میلی متر و میانگین عرض آن 14/3 میلی متر تعیین گردید. در بررسی مقاطع بافتی مشاهده شد که غده از چندین لوب تشکیل شده است که از سطح خارجی توسط کپسول نازکی احاطه شده اند. از کپسول، رشته های ظریفی به نام سپتا به داخل غده کشیده می شود و آن را به لوبول هایی تقسیم می کند. شکل واحد های ترشحی آن از نوع لوله ای آلوئولی مرکب و بافت پوششی دیوارۀ آن از نوع استوانه ای ساده می باشد. ترشحات، غدۀ هاردرین با رنگ آمیزی های پاس و آلسین بلو، رنگ آمیزی شد که در نتایج به دلیل وجود موسین های اسیدی در سلول های واحد ترشحی و مجاری این ساختارها به شدت به رنگ آلسین بلو واکنش مثبت نشان دادند ولی بعلت ناچیز بودن ترکیبات موسینی خنثی، سلول های ترشحی این غده به رنگ آمیزی پاس واکنش ضعیفی را نشان دادند و ترشحات بصورت ارغوانی کم رنگ مشاهده شدند.

Histomorphology and histochemical study of harderian gland in Cockatiel (Nymphicus hollandicus)

The anatomical structure of the eye, the type of lacrimal glands and its secretions can be important in the absence of some damage to the eye. Five healthy male cockatiels were used in this research.

After dissecting the specimens and detrmining the location of the gland,tissue sections were prepared using usual tissue preparation methods and stained with hematoxylin-eosin, PAS, Alcian blue and toluidine blue. The results showed that Harderin's gland in the cockatiel is triangular and located in the inner ventral surface of the eyeball. Its average length was 4.5 mm and its average width was 3.14 mm. In tissue sections, it was observed that this gland consists of several lobes that are surrounded by a thin capsule with fine threads called septa extend into the gland and divide it into lobules. The shape of the secretory units of Harderin gland was compound tubular alveolar type and the covering tissue of its wall was simple columnar type. The secretions of Harderin gland were stained with PAS and Alcian blue stains, which showed a significant response to Alciain blue staining in the results of secretory cells of the gland and due to the presence of acidic mucins in the cells of the secretory unit and ducts of these structures. They showed a positive reaction to this stain, but due to the small amount of neutral mucin compounds, the secretory cells of this gland showed a weak reaction to PAS staining and the secretions were observed as pale purple.
