چکیده
دوزیستان بیش از ۳۰۰ میلیون سال است بر روی کره زمین زندگی میکنند. به نظر برخی محققان دو گونه از جنس Pelophylax Fitzinger, 1843 در ایران وجود دارد. در این تحقیق ساختار پوست 48 قورباغه آبی شامات Pelophylax bedriagae (Camerano, 1882) از چهار جمعیت مختلف در شمال ایران مورد مطالعه قرار گرفت. نمونهگیری از مناطق گرگان، آلاگل، ساری و تنکابن، این نمونهها به آزمایشگاه منتقل و بخشی از پوست سطح پشتی و شکمی آنها جدا شد. پس از انجام مراحل آمادهسازی بافت و رنگآمیزی هماتوکسیلین- ائوزین، چهار ویژگی پوست آنها شامل ضخامت اپیدرم، ضخامت درم، ضخامت کل پوست و تعداد غدد موکوسی در یک میلیمتر مربع از پوست، در زیر میکروسکوپ نوری بررسی و دادههای بهدست آمده آنالیز شد. نتایج آزمونهای t و Mann-Whitney برای همه صفات بین گروههای نر و ماده تفاوت معنیداری نشان نداد. نتایج آزمونهای پارامتری ANOVA و ناپارامتری Kruskal-Wallis نشان داد که میان جمعیتهای مورد بررسی در همه صفات تفاوت معنیدار وجود دارد. آزمون آنالیز واریانسهای چندمتغیره (MANOVA) نشان داد که جدایی جمعیتها کاملاً معنیدار است. نتایج حاصل از تحلیل خوشهای، نمونهها را بر اساس فاصله به دو گروه تقسیم کرد که گروه اول شامل نمونههای گرگان و ساری و گروه دوم شامل نمونههای آلاگل و تنکابن بود. نتایج تحلیل کانونیکال نشان داد که 58/89 ٪ نمونهها در دستهبندی خودشان قرار میگیرند. به طور کلی، این تحقیق نشان داد که میتوان از صفات پوستی در تحلیلهای تشخیصی استفاده کرده و این گروه از جانوران را از هم جدا کرد.

Histological structure of skin in Pelophylax bedriagae and its relationship with ecological factors in Golestan & Mazandaran provinces

Amphibians have lived on Earth for more than 300 million years. Two species of Pelophylax have been reported in Iran, which are Pelophylax bedriagae and Pelophylax gigas. In the present study, the skin structure of 48 water frogs (Pelophylax bedriagae) in four different regions has been investigated. After taking specimens from Gorgan, Alagol, Sari and Tonekabon regions, these specimens were transferred to the laboratory and a part of their dorsal and ventral surface skin was separated. After tissue preparation and Hematoxylin-Eosin staining, four skin characteristics, including epidermis thickness, dermis thickness, total skin thickness, and the number of mucous glands in one mm2 of skin, were examined under a light microscope and the obtained data were analyzed. The results of T and Mann-Whitney tests for all traits did not show significant differences between male and female groups. The results of parametric ANOVA and non-parametric Kruskal-Wallis tests showed that there is a significant difference between the investigated populations in all traits. The multivariate analysis of variance (MANOVA) test showed that the separation of populations is completely significant. The results of the cluster analysis divided the specimens into two groups based on the distance, the first group included the samples of Gorgan and Sari and the second group included the specimens of Alagol and Tonekabon. The results of the canonical analysis showed that 89.58% of the specimens fall into their categories. In general, this research showed that skin traits can be used in diagnostic analyzes and separate this group of animals.
