تبارزایی مولکولی گاوماهیان ایران بر اساس نشانگرهای میتوکندریایی و هسته ای

چکیده

گاوماهیان متنوع ترین خانواده ماهیان استخوانی بوده و تقریبا 1915 گونه متعلق به 263 جنس را در خود جای داده است. این خانواده از پنج زیرخانواده شامل Sicydiinae، Gobionellinae، Oxudercinae، Amblyopinae و Gobiinae تشکیل شده‌اند. تاکنون 24 گونه گاوماهی متعلق به 19 جنس از آب های ایرانی خلیج فارس و دریای عمان گزارش شده‌اند.. هدف مطالعه حاضر بررسی روابط تبارزایی گاوماهیان ایران با استفاده از 64 توالی (نشانگرهای COI، Rag1 و Rho) مربوط به 21 گونه است. در این مطالعه، نشانگرهای مولکولی سه گونه Bathygobius meggitti (چابهار، ژن ردوپسین)، Stonogobiops nematodes و Valenciennea strigata (جزیره قشم، ژن COI) برای اولین بار تعیین توالی شده اند. گاوماهیان ایران متعلق به دودمان های مولکولی متنوعی هستند که عبارتند از: Periophthalmus-lineage، Gobiopsis-lineage، Glossogobius-lineage، Cryptocentrus-lineage، Gobiodon-lineage، Valenciennea-lineage. تعیین توالی ژن های هسته‌ای تمام گونه‌های شناسایی شده در جنوب ایران و حوضه دریای خزر می‌تواند تصویر بهتری از روابط تبارزایی تاکسون‌های این خانواده نشان دهد.

چکیده [English]

Gobiidae is the most species-rich family of teleost fishes, with approximately 1915 valid species belonging to 263 genera. This family is composed of five subfamilies, including Sicydiinae, Gobionellinae, Oxudercinae, Amblyopinae, Gobiinae. 24 species from 19 genera of Gobiidae have been reported in Iranian waters of the Persian Gulf and the Gulf of Oman. The aim of the present study is to survey phylogenetic relationships of Gobiidae representatives from Iran using 64 sequences (Rhodopsin, Rag1 and COI markers) belong to 21 species. In this study, specimens from Bathygobius meggitti (Chabahar, sequenced for Rhodopsin), Stonogobiops nematodes, and Valenciennea strigata (Qeshm Island, sequenced for COI) have been examined for the first time. Gobiidae members recorded from Iran are classified in diverse molecular lineages including, Periophthalmus-lineage, Gobiopsis-lineage, Glossogobius-lineage, Cryptocentrus-lineage, Gobiodon-lineage and Valenciennea-lineage. Sequencing nuclear markers of all recorded species from the Iranian Southern water bodies and the Caspian Sea basin could determine more detail of molecular phylogenetic relationships of this family.

مقدمه

 

گاوماهیان متنوع­ترین خانواده ماهیان استخوانی و تقریباً 1943 گونه متعلق به 258 جنس را در خود جای داده است (10). این ماهیان معمولاً کوچک و ساکن آب های شور هستند، باله­های پشتی خاردار، و باله های لگنی معمولاً به یکدیگر متصل شده و تشکیل یک دیسک را میدهند. الگوهای رنگی متنوعی در این ماهیان مشاهده می­شوند که معمولاً برای استتار جانور مفید هستند و مشاهده اعضای این خانواده را در محیط زیست شان دشوار و در برخی مواقع غیرممکن می کند (41). این خانواده از پنج زیرخانواده شامل Sicydiinae، Gobionellinae، Oxudercinae، Amblyopinae و Gobiinae تشکیل شده­ است (48). رده­بندی های متفاوتی براساس صفات ریخت شناسی برای راسته Gobiiformes Günther, 1880 ارائه شده است که این رده­بندی­ها در مورد تعداد خانواده ها و زیرخانواده ها با یکدیگر توافق ندارند (3، 4، 5، 15، 17، 18، 24 و 28). مطالعات تبارزایی مولکولی، دودمان های گاوماهی شکلان عمده را براساس نشانگرهای میتوکندریایی و هسته­ای مورد بررسی قرار داده­اند (1، 2، 23، 44، 45و 47). در یکی از اولین مطالعات (39) سه ژن میتوکندریایی ND1، ND2 و COI مورد استفاده قرار گرفته و تک نیایی خانواده های تعریف شده با صفات ریختی تحت آزمون مولکولی قرارگرفتند. نتایج این مطالعه نشان داد که خانواده Xenisthmida درون Eleotridae و همچنین Eleotridae باتوجه به Gobiidae، Microdesmidae، Ptereleotridae، Kraemeriidae و Schindleriidae پیراتبار است. به علاوه، کلاد Gobionellinae باتوجه به Kraemeriidae، Sicydiinae، Oxudercinae و Amblyopinae پیراتبار است. مطالعه مذکور نتیجه گیری می کند که کاهش صفات ریختی در بسیاری از گونه های گاوماهیان چندین بار و به طور مستقل از یکدیگر تحت فشارهای بوم شناختی رخ داده است. مطالعه بعدی (44) با استفاده از چهار ژن میتوکندریایی (ND1، ND2، COI و cytb) تبارزایی دودمان Gobioidei را بررسی کرد. در این مطالعه خانواده های Rhyacichthyidae و Odontobutidae تک نیا ولی خانواده های Eleotridae، Xenisthmidae و  Gobiidae پیراتبار تشخیص داده شدند. بررسی دوباره خانواده Gobiidae (40) با استفاده از چهار ژن میتوکندریایی (ND1، ND2، COI و cytb) منجر به معرفی خانواده Gobionellidae گردید که اعضای آن شامل چهار زیرخانواده Gobionellinae، Oxudercinae، Amblyopinae و Sicydiinae می باشند و اعضای زیرخانواده Gobiinae در یک خانواده با نام قدیمی Gobiidae باقی ماندند. گردآوری داده­های مولکولی و رسم درخت تبارزایی مربوطه (1) نیز تک نیایی زیرخانواده های Gobionellinae، Amblyopinae و Oxudercinae را تایید نکرده است. در تحلیل مولکولی بعدی (45) نشانگرهای مولکولی میتوکندریایی (ND1، ND2 و COI) و هسته ای (RAG2، Rhodopsinو RNF213) برای مطالعه تبارزایی زیرخانواده Gobiinae مورد استفاده قرارگرفتند و 13 دودمان شناسایی شده است که از مهم ترین آن ها می توان Lagoon gobies، Silt shrimp gobies، Flapheaded gobies و Burrowing paired gobies را نام برد. در تحلیل دیگری (47) با استفاده از دو ژن هسته ای (Rag1 و Rhodopsin) تبارزایی پنج زیرخانواده متعلق به خانواده Gobiidae بررسی شد و دو دودمان اصلی شامل دودمان Gobiidae (sensu stricto of Thacker 2009) و دودمان Gobionellidae (sensu Thacker 2009) شناسایی شدند. یکی از کامل ترین مطالعات مولکولی روی دودمان Gobiodei با استفاده از دو ژن میتوکندریایی (rRNA و cytb) و سه ژن هسته ای (RAG1، zic1 و sreb2) انجام گرفت (2). نتایج این بررسی نشان داد که Rhyacichthydae و Odontobutidae گروه خواهری تمام دودمان های دیگر هستند و همچنین در کلاد دیگری خانواده های Butidae و Gobiidae به صورت دو دودمان خواهری قرار گرفته­اند. درون خانواده Gobiidae دو کلاد gobiine-like و gobionelline-like تشخیص داده شده اند که هریک از این کلادها دارای تعدادی از دودمان های شناسایی شده اند که الزاما منطبق بر ویژگی های ریختی و یا سازگاری های بوم شناختی نیستند. 

تاکنون 24 گونه متعلق به 19 جنس از گاوماهیان از آب های ایرانی خلیج فارس و دریای عمان گزارش شده­اند (6، 7، 11، 12، 14، 21، 33، 34، 35، 36و 37)، اگرچه 61 گونه متعلق به 35 جنس از این خانواده از آب های ساحلی کشور عمان گزارش شده است (31). تفاوت نسبتاً زیاد در تعداد گونه های گزارش شده از این مناطق می تواند نشان دهنده حضور گونه های گزارش نشده و یا ناشناخته در بخش های شمالی خلیج فارس و دریای عمان باشد. تبارزایی مولکولی سه گونه گلخورک آب های سواحل جنوبی ایران (Boleophthalmus dussumieri, Periophthalmus waltoni, Scartelaos tenuis) با استفاده از دو ژن میتوکندریایی (COI و Cyt b) و یک ژن هسته­ای (Rag1) مطالعه شده­اند (13و 29). روابط تبارزایی هفت گونه گوبی خلیج فارس و دریای عمان با استفاده از دو ژن هسته ای (Rho و Rag1) بررسی شد و نشان داد که می توان 2 کلاد و 4 زیرکلاد را تعریف کرد (14). چهار گونه از منطقه زیرجزرومدی جزیره قشم متعلق به دو جنس از گاوماهی­ها (Gobiodon citrinus, Valenciennea puellaris, V. sexguttata, V. strigata) با استفاده از دو نشانگر میتوکندریایی و هسته ای (Rho و COI) مورد مطالعه قرار گرفته اند (25). بررسی صفات ریخت شناسی و مولکولی موجب شناسایی گونه جدید Silhouettea ghazalae از جزیره قشم شده است، همچنین 12 گونه از گاوماهیان آب های سواحل جنوبی ایران با استفاده از ژن میتوکندریایی COI مورد مطالعه و روابط تبارزایی آن ها مشخص گردید (21). 

هدف از مطالعه حاضر، بررسی روابط تبارزایی گاوماهیان ایران (20 گونه از گاوماهیان گزارش شده از جنوب ایران و یک گونه از گاوماهیان حوضه دریای خزر) با استفاده از داده های سه ژن تعیین توالی شده در مطالعه حاضر و داده های موجود در بانک ژنی مربوط به یک ژن میتوکندریایی (COI) و دو ژن هسته­ای (Rho و Rag1) می باشد.

مواد و روشها

نمونه­های ماهی از سه گونه Bathygobius meggitti (چابهار 25°20'48.8" N 60°35'47.6" E)، Stonogobiops nematodes (جزیره قشم 26°55'38.8" N 56°14'57.7" E) و Valenciennea strigata (جزیره قشم) جمع آوری شدند (شکل1). B. meggitti از منطقه بین جزرو مدی و
S. nematodes و V. strigata از منطقه زیرجزرو مدی جمع آوری شدند. نحوه تکثیر ژن ردوپسین برای گونه
B. meggitti در مقاله Mohammadi and Ghanbarifardi, 2020 و نحوه تکثیر ژن  COI برای دو گونه
S. nematodes و V. strigata در مقاله Kovačić et al. (2020) توضیح داده شده است. هر سه نمونه در موزه جانورشناسی دانشگاه سیستان و بلوچستان (ZMUSB) نگهداری می­شوند. سه توالی حاصل در بانک ژنی با شماره­های بازیابی MZ427500، MZ379280، MZ379281 قرارگرفتند. 61 توالی مربوط به 21 گونه گاوماهی از طریق بانک ژنی در دسترس هستند (جدول 1) و برای رسم درخت تبارزایی گاوماهیان ایران استفاده شده­اند. لازم به ذکر است اکثر توالی های مورد استفاده از نمونه­های ایرانی استخراج شده­اند و در مطالعات مختلف مورد استفاده قرار گرفته­اند (13، 14، 21، 25 و 29).

Kurtus gulliveri با توجه به مطالعه مولکولی گاوماهیان به عنوان برون گروه انتخاب شد (2). درخت های تبارزایی با روش بیشینه درست نمایی (ML) (9) و بیژین (BI) (19) رسم شدند. تحلیل ML با استفاده از RAxML v.8.1.24 (38) با 1000 تکرار بوت استرپ و تحلیل BI با استفاده از MrBayes v.3.2.6 (32) با 10 میلیون نسل تکرار انجام گرفتند. برای توالی های مورد استفاده در این مطالعه بهترین مدل جایگزینی نوکلئوتیدی با استفاده از Partition Finder version 2 (22) مشخص شد و مدل جایگزینی GTRGAMMAI برای تحلیل های تبارزایی استفاده شد.

 

 

 

شکل2- روابط تبارزایی خانواده گاوماهیان حاصل از روش بیشینه درست­نمایی شامل دو ژن هسته­ای (Rag1 و Rho) و یک ژن میتوکندریایی (COI) (مجموعا 2483 جفت باز). اعداد به ترتیب نشان دهنده بوت استرپ حاصل از آنالیز بیشینه درست­­نمایی و احتمال پسین از تحلیل بیژین می­باشد. در این مطالعه، نشانگرهای مولکولی سه گونه Bathygobius meggitti (ردوپسین)، Stonogobiops nematodes و Valenciennea strigata (COI) برای اولین بار تعیین توالی شده­اند که با رنگ قرمز نشان داده شده­اند. رنگ­های مختلف کلاد­ها به ترتیب از بالا به پایین درخت نشان دهنده خوشه­های 1 تا 6 می‍باشند که در متن توضیح داده شده اند. آرایه های خوشه اول متعلق به زیرخانواده Oxudercinae و خوشه های دو تا شش متعلق به زیرخانواده Gobiinae می­باشند.

 

 

نتایج

نشانگر هسته­ای ردوپسین برای گونه Bathygobius meggitti برای اولین بار تعیین توالی گردید؛ همچنین نشانگر COI برای دو گونه Stonogobiops nematodes و Valenciennea strigata صید شده از جزیره قشم تعیین توالی شد. داده­های ترکیبی (جدول1) شامل 2483  جفت باز (Rag1: 1162, COI: 651, Rhodopsin: 670) که از این تعداد 775 جفت باز متغییر (Rag1: 349, COI: 263, Rhodopsin: 163) و 658 جفت باز دارای اطلاعات پاسیمونی (Rag1: 271, COI: 249, Rhodopsin: 138) بودند. هر دو درخت بیشینه درست­نمایی و بیژین شباهت زیادی به یکدیگر دارند، البته در طول شاخه­ها و میزان حمایت با یکدیگر تفاوت­هایی دارند.

درخت تبارزایی شکل 2 را می توان متشکل از 6 خوشه اصلی دانست. خوشه اول شامل سه گونه گلخورک
P. waltoni، B. dussumieri، S. tenuis متعلق به زیرخانواده Oxudercinae، خوشه دوم متشکل از
 B. meggitti، B. cocosensis، S. nematodes،
 C. cyanotaenia، C. arabicus، F. reiche، I. ornatus، A. dayi، خوش سوم شامل P. scapulopunctatus، Silhouetta ghazalae، خوشه چهارم در برگیرنده
 A. ocellata، V. strigatta، V. sexguttata،
 V.  puellaris،G. citrinus ، خوشه پنجم شامل
 C. admsoni، C. tessellatus و خوشه ششم دارای
P. iranicus هستند. هرکدام از خوشه ها روی شکل 2 با رنگ های متفاوتی نشان داده شده اند. آرایه های خوشه های 2 تا 6 همگی متعلق به زیرخانواده Gobiinae می‍باشند.    

بحث

میزان­های جهش در ژن­های هسته­ای کمتر از ژن­های میتوکندریایی هستند؛ بنابراین ژن­های هسته­ای که دارای کپی­های ژنی کد کننده پروتئینی محدودی می­باشند، روش­های مناسبی را برای استنباط­ تبارزایی فراهم می­کنند (8، 16 و 26) و به همین دلیل است که تحلیل های مولکولی گاوماهیان در سال های اخیر از نشانگرهای هسته ای استفاده کرده اند (2، 14، 45، 47).

خوشه اول در این مطالعه از سه گونه گلخورک
(P. waltoni، B. dussumieri، S. tenuis) تشکیل شده است که در سواحل گلی و جنگل­های حرای جنوب ایران پراکنش دارند و جز دودمان Periophthalmus-lineage محسوب می­شوند (2، 13، 14، 29). برخی از مطالعات مولکولی دلایلی را ارائه می­دهند که بر طبق آن­ها خانواده گاوماهیان به دو کلاد عمده به نام­های خانواده Oxudercidae (Gobionellidae) و Gobiidae تقسیم بندی می­شوند. خانواده Oxudercidae شامل چهار زیرخانواده Gobionellinae، Oxudercinae، Amblyopinae و Sicydiinae می­باشد که از 86 جنس تشکیل شده و توسط برخی صفات ریخت شناسی حمایت می­شود (2، 27، 39، 40، 41 و 45). رده­بندی­های عمده ماهی­شناسی هنوز هم خانواده گاوماهیان را به عنوان یک خانواده با پنج زیرخانواده معتبر می­دانند (10) یا اینکه از واژه زیرخانواده استفاده نمی کنند و تمام اعضای مورد مطالعه را در خانواده Gobiidae قرار می دهند (2)، بنابراین مطالعه حاضر هم از رده بندی خانواده گاوماهیان با پنج زیرخانواده استفاده می‍شود و خانواده Oxudercidae (Gobionellidae) را به عنوان یک آرایه بکار نمی برد. خوشه دوم شباهت­هایی به سه دودمان Glossogobius-lineage (BathygobiusCryptocentrus-lineage (Cryptocentrus و Cryptocentroides) و Gobiopsis-lineage (Favonigobius، Istigobius و Acentrogobius) در مطالعه آگورتا و همکاران (2013) (2) دارد که به صورت خوشه های خواهری در کنار یکدیگر قرار گرفته­اند. لازم به ذکر است که در دودمان Cryptocentrus-lineage (2) جنس Stonogobiops در نظر گرفته نشده است ولی در دودمان Cryptocentrus-lineage مطالعات تَکِر (2015) و قنبری فردی و لکزیان (2019) جنس Stonogobiops نیز جز دودمان مذکور قرار می گیرد. Stonogobiops و Cryptocentrus از لحاظ شکل غشای آبششی و ساختارهای استخوان های تیغه ای و استخوان های کامی دارای خویشاوندی نزدیکی هستند (46) که این خویشاوندی در درخت تبارزایی مطالعه حاضر (شکل 2) نیز نشان داده شده است. تَکِر و روج (2011) سیزده دودمان گاوماهیان را براساس داده­های مولکولی و ریخت شناسی تعریف کرده­اند که دودمان گوبی­های تالابی (Gobiopsis-lineage) جنس­های Acentrogobius، Istigobius، Favonigobius، Silhouettea و Aulopareia را در برمی­گیرد؛ مطالعه حاضر نیز تا حدی تبارزایی مشابهی را ارائه می دهد. در مطالعه حاضر فقط ژن COI برای سه گونه Silhouettea ghazalae، Aulopareia ocellata و Palutrus scapulopunctatus مورد استفاده قرارگرفت و به نظر می­رسد برای تعیین جایگاه دقیق­تر تبارزایی این سه گونه بایستی ژن­های بیشتری تعیین توالی و مورد استفاده قرار گیرند. دودمان های Gobiodon-lineage و Valenciennea-lineage (خوشه چهارم) از جمله دودمان­های با خویشاوندی تقریبا نزدیک در خانواده گاوماهیان هستند (2 و 43) البته خویشاوندی Valenciennea با جنس­های Signigobius و Amblygobius و همچنین خویشاوندی Gobiodon با Paragobiodon با استفاده از مارکرهای میتوکندریایی و هسته ای مشخص شده است (25). Coryogalops و Ponticola هر دو در دودمان Gobius-lineage قرار می‍گیرند (2)، لازم به ذکر است که جنس Coryogalops بیشتر در سواحل آفریقا و دریای سرخ پراکنش دارد (20) در حالی که پنج گونه Ponticola در آب­های شیرین حوضه آبریز Ponto-Caspian پراکنش دارند (49).

برای مطالعه دقیق تر تبارزایی گاوماهیان ایران بایستی تمام گونه­ها برای نشانگرهای هسته­ای و میتوکندریایی یکسانی تعیین توالی شوند، همچنین جنس هایی مانند Neogobius و Hetereleotris به علت نبودن توالی­های مربوط به نمونه­های ایران در این مطالعه بررسی نشدند که می­توانند هدف مطالعات آینده باشند.
